The Inhibitory Effect of Phellodendri Cortex and Clematidis Radix Aqua - Acupuncture to IL - 1¥â in LPS induced Arthritis.



Abstract
Objective : Lipoplysaccharide(LPS) of E.coli¢¥s has been considered to promote the inflammation and the resulting inflammation was thought to be attributed to production of cytokines, such as interlecukin-1(IL-1), tumour necrosis factor ¥á(TNF-¥á) and other inflammatory mediators.
Injection of Lipopolysaccharide(LPS) into mouse knee joints provoked leukocyte infiltration and loss of proteoglycan(PG) from the cartilage.
IL-1 was potentially an important inflammatory mediator because of its wide range of effects and it contributed to the destruction of cartilage in inflammatory disease, especially rheumatoid arthritis(RA).
The aim of the present study was to examine the role of IL-1¥â in LPS-induced arthritis in mouse knee joints.
Furthermore it was undertaken to investigate the anti-inflammatory effects of Phellodendri Cortex and Clematidis Radix aqua-acupuncture in LPS-induced arthritis of mouse knee joints.
Materials and Methods : The experimental model of arthritis was induced by injection of 10¥ìg LPS in mice weighing 18¡­30g.
The Phellodendri Cortex and Clematidis Radix aqua-acupuncture were injected into Du¡Çbi(ST-35) and blank locus in mice.
The measurement have been performed as follows.
(1) MTT test of the Phellodendri Cortex and Clematidis Radix water extract, (2) Reproduction of arthritis by hrIL-1¥â, (3) Leukocyte infiltration in mouse monocyte by LPS, (4) Production of IL-1¥â and TNF ¥á in SF by SPS using ELISA, (5) Leukocyte infiltration and the loss of PG by the Phellodendri Cortex and Clematidis Radix aqua-acupuncture.
Results : the obtained results are summarized as follows;
1. Production of IL-1¥â in mouse monocyte induced by LPS injection was depended upon LPS concentration.
2. IL-1¥â and TNF in mouse synovial fluids were overproducted by LPS stimulation.
3. Leukocyte infiltration in mouse joint increased when recombinant IL-1¥âwas injected.
4. In mouse monocyte, both Phellodendri Cortex and Clematidis Radix water extracts did not show any cellualr toxicity by 200¥ìg/§¢.
5. Production of IL-1¥â in mouse monocyte was inhibited by the injection of Phellodendri Cortex and Clematidis Radix water extracts.
6. The Phellodendri Cortex aqua-acupuncture inhibited leukocyte infiltration in LPS-induced arthritis.
7. The Phellodendri Cortex aqua-acupuncture pretratment inhibited leukocyte infiltration in LPS-induced arthritis.
8. The Phellodendri Cortex aqua-acupuncture pretratment supressed loss of proteoglycan in LPS-induced arthritis.
9. The Phellodendri Cortex aqua-acupuncture recovered proteoglycan content in LPS-induced arthritis.
10. The Phellodendri Cortex aqua-acupuncture used affected to the transcriptional regulation level of the IL-1¥â gene expression in in-vivo experiment.
Conclusion : According to the above results, it was concluded that IL-1¥â acts as a key mediator in LPS-arthritis and the Phellodendri Cortex aqua-acupuncture was highly effective for anti-inflammation reaction.
